Sample Question 1

Assessment Focus: Understanding how inconsistent definitions affect transparency and reproducibility in synthetic-data research
—————————————–

Two research teams describe their medical imaging datasets as “synthetic.” Team A creates new chest radiographs using a purpose-built generative model, while Team B only rotates and crops existing patient images without clearly reporting its procedures.

According to the article, which action would most directly improve the transparency and reproducibility of these studies?

  1. Accept both datasets as equivalent because all digitally modified images are synthetic.
  2. Require each team to describe its data-generation method, source data, intended task, model settings, and post-processing procedures using standardised reporting guidelines.
  3. Evaluate only whether the images look realistic to nonmedical observers.
  4. Exclude all modified images because synthetic data cannot be used in medical research.
  5. Classify a dataset as synthetic only when no original patient data were used at any stage.

Correct Answer: 2

Example: Reasoning / Explanation

The term “synthetic data” may be interpreted differently across studies. Standardised reporting would allow researchers to understand how each dataset was produced and determine whether the findings can be reproduced or compared fairly.

Example: Academic Principle / Supporting Evidence

The article explains that the absence of a universally accepted definition can reduce research transparency and reproducibility. It also recommends disclosing source datasets, generation conditions, model settings, and post-processing steps.

Sample Question 2

Assessment Focus: Interpreting the difference between crude and age-standardised cardiovascular mortality rates
—————————————–

The article reports that Japan had a higher crude cardiovascular disease mortality rate than South Korea, but Japan had a lower age-standardised mortality rate.

Which interpretation is most appropriate?

  1. Age standardisation proves that cardiovascular disease does not occur in older Japanese adults.
  2. Japan’s older population structure may increase its crude mortality rate, while age adjustment suggests a lower underlying age-adjusted mortality rate than South Korea.
  3. South Korea must have more elderly people because its crude mortality rate is lower.
  4. Crude mortality rates remove the effect of age, whereas age-standardised rates do not.
  5. The two measures are contradictory, so neither can be used to compare populations.

Correct Answer: 2

Example: Reasoning / Explanation

Crude mortality is influenced by the age distribution of a population. A country with more older adults may have a higher crude death rate even when its age-adjusted cardiovascular mortality is comparatively lower.

Example: Academic Principle / Supporting Evidence

Age standardisation allows populations with different age structures to be compared more fairly. The figure shows that Japan’s crude rate is relatively high, but its age-standardised rate is lower than South Korea’s.

—————————————–

Sample Question 1

Assessment Focus: Designing clinically meaningful validation for generative AI used in surgical planning
—————————————–

A generative model creates predicted postoperative hip radiographs from preoperative images. Specialist surgeons rate the generated images as highly realistic and anatomically accurate. A hospital now proposes using the model to support patient-specific surgical planning.

Which next step would provide the strongest evidence for responsible clinical implementation?

  1. Measure only the time required to generate each image.
  2. Ask the model developers whether the generated images appear medically reasonable.
  3. Conduct a prospective, multi-centre study comparing predicted images with actual postoperative outcomes and evaluating whether the model improves surgical planning without increasing clinical errors.
  4. Use the model immediately because expert ratings prove that its predictions are accurate for every patient.
  5. Replace actual postoperative imaging with generated images in all future cases.

Correct Answer: 3

Example: Reasoning / Explanation

High visual and anatomical ratings do not prove that the model accurately predicts real postoperative outcomes or improves clinical decisions. The model should be tested prospectively in independent populations and evaluated for both benefit and possible harm.

Example: Academic Principle / Supporting Evidence

The article distinguishes perceptual realism from clinical utility. Synthetic medical images require task-specific validation, evaluation by multiple experts, independent testing, and evidence that clinical performance remains reliable.

—————————————–

Sample Question 2

Assessment Focus: Redesigning a cardiovascular cohort to reduce selection bias and improve subgroup-specific risk prediction
—————————————–

Researchers plan to develop an ASCVD risk calculator for all East Asian Americans. Their initial dataset contains only English-speaking patients from one private health-care system, and all Chinese, Japanese, and Korean participants are recorded under one category labelled “Asian.”

Which redesign best addresses the major limitations identified in the article?

  1. Increase the number of participants from the same private health-care system without changing the recruitment method.
  2. Combine East Asian participants with South Asian and Southeast Asian participants to create a larger sample.
  3. Recruit participants from multiple regions and health-care settings, provide native-language participation options, oversample under-represented subgroups, and record country of origin, immigration history, and generational status.
  4. Remove environmental and socioeconomic information because ancestry alone determines cardiovascular risk.
  5. Apply a Western risk calculator to the original sample without further validation.

Correct Answer: 3

Example: Reasoning / Explanation

The original design may exclude people with limited English proficiency and may not represent individuals outside a private health-care system. Combining all East Asian participants into one group can also hide important differences among Chinese, Japanese, and Korean populations.

Example: Academic Principle / Supporting Evidence

The article recommends disaggregated subgroup data, broader participation, native-language recruitment, and consideration of acculturation and environmental factors. These measures improve representativeness and support more accurate risk-model development and validation.

Sample Question 1

Assessment Focus: Comparing land-use strategies for sustainable agricultural development
—————————————–

The article discusses two land-use approaches:

  • Land sparing: producing food intensively on some land while protecting other areas for conservation.
  • Land sharing: integrating agricultural production with habitats and biodiversity within the same landscape.

Why does the article suggest that combining these two approaches may be more suitable than relying entirely on only one approach?

  1. Because combining them eliminates the need to evaluate agricultural productivity.
  2. Because land sparing always increases biodiversity, while land sharing always increases crop yield.
  3. Because a combined strategy can protect important natural areas while allowing biodiversity and agricultural production to coexist in appropriate parts of the landscape.
  4. Because the two approaches are scientifically identical and differ only in terminology.
  5. Because combining them allows all agricultural land to be converted into protected forests.

Correct Answer: 3.

Answer Rationale:
Land sparing and land sharing address different parts of the conservation–production challenge. Protecting some natural areas may conserve sensitive ecosystems, while integrating biodiversity into farmland can support ecosystem services and landscape connectivity. The article therefore indicates that a combination may provide a more balanced path toward long-term conservation and agricultural production.

Reasoning / Explanation

Combining the two approaches is more appropriate because each approach addresses a different part of the conflict between agricultural production and conservation. Land sparing can protect ecologically sensitive areas, while land sharing allows biodiversity and farming to coexist within productive landscapes. Therefore, using both approaches can produce a more balanced result than relying entirely on either one.

Academic Principle / Supporting Evidence

The relevant academic principle is integrated land-use planning. The article defines land sparing as preserving some land while intensively using other areas, whereas land sharing combines agricultural activities with the natural environment. It states that combining the two approaches may provide an optimal pathway for long-term conservation and sustainable agricultural production.

—————————————–

Sample Question 2

Assessment Focus: Relating electrospun scaffold structure to biological and mechanical functions
—————————————–

Electrospinning can produce hydrogel-based fibers with diameters ranging from the nanometer to micrometer scale. The article reports that electrospun nanofiber networks may improve water absorption, mechanical properties, and cell proliferation.

Which explanation best accounts for these combined effects?

  1. Nanofibers completely prevent water and nutrients from entering the scaffold.
  2. Their small diameter and high surface area can create an extracellular-matrix-like structure that supports cell attachment while reinforcing the hydrogel network.
  3. Electrospinning converts all reversible crosslinks into permanent metallic bonds.
  4. Smaller fibers always eliminate the need to test scaffold strength and biocompatibility.
  5. Nanofiber networks support cells only because they cause the hydrogel to degrade immediately.

Correct Answer: 2.

Answer Rationale:
Electrospun fibers can produce porous structures resembling the extracellular matrix. Their high surface area provides additional sites for cell attachment, while the fiber network may reinforce the hydrogel and improve water interaction and molecular transport.

Reasoning / Explanation

The nanofiber network is the best explanation because its small fibers create a porous structure with a large surface area. This provides more sites for cells to attach and can distribute mechanical forces through the scaffold. The same network can also interact with water and support molecular transport, explaining the improvement in both biological and material performance.

Academic Principle / Supporting Evidence

The relevant principle is the structure–property–function relationship in biomaterials. Electrospinning produces fibers at nanometer-to-micrometer scales, creating structures resembling the extracellular matrix. The article reports that electrospun nanofibers improved water absorption, mechanical properties, cell survival, and rapid shape recovery while maintaining their chemical structure.

—————————————–

Sample Question 1

Assessment Focus: Applying carbon-management principles to agricultural development in peatland areas
—————————————–

A province plans to expand crop production into a drained peatland area. The drained land is releasing large amounts of carbon dioxide, but the province must also maintain food production and protect farmers’ incomes.

Which strategy is most consistent with the evidence and systems approach presented in the article?

  1. Continue draining the entire peatland and compensate for the emissions by increasing fertilizer use.
  2. Burn the remaining peat vegetation before cultivation to reduce future waste-management costs.
  3. Ignore peatland emissions because only methane and nitrous oxide are relevant to agriculture.
  4. Restore the most carbon-intensive degraded peatland areas, improve productivity on suitable existing farmland, monitor soil-carbon changes, and provide transition support for affected farmers.
  5. Convert all agricultural land in the province into peatland without assessing ecological or economic conditions.

Correct Answer: 4.

Answer Rationale:
The article identifies peatlands as major natural carbon stores and damaged peatlands as important greenhouse-gas sources. A suitable strategy should therefore reduce emissions from degraded peat while maintaining agricultural productivity through better management of appropriate farmland. Support for farmers is also needed to avoid transferring environmental costs into economic hardship.

Reasoning / Explanation

Restoring the highest-emitting peatland areas while improving production on existing suitable farmland addresses both climate and food-security objectives. Continued drainage would release more stored carbon, whereas stopping all farming could harm food production and farmers’ incomes. The selected strategy reduces environmental damage while maintaining production and supporting affected communities.

Academic Principle / Supporting Evidence

The relevant principles are carbon sequestration, ecosystem restoration, and sustainable intensification. Peatlands store exceptionally large amounts of terrestrial carbon, but damaged or drained peatlands become major greenhouse-gas sources. The article reports that damaged peatlands account for more than 5% of total human carbon-dioxide emissions and identifies peatland restoration as an effective natural carbon-storage strategy.

—————————————–

Sample Question 2

Assessment Focus: Selecting a fabrication mechanism for an injectable and biologically active hydrogel
—————————————–

Researchers need to develop an injectable shape memory hydrogel containing a temperature-sensitive therapeutic molecule. The preparation method should use mild conditions, permit self-healing, and avoid permanent damage to the therapeutic molecule.

Which approach is most appropriate?

  1. Use self-assembly driven by reversible non-covalent interactions such as hydrogen bonding, hydrophobic interactions, and π–π stacking, followed by testing stability under physiological conditions.
  2. Apply continuous extreme heat until all polymer chains form irreversible bonds.
  3. Remove all intermolecular interactions so that the hydrogel cannot form a three-dimensional network.
  4. Use only a rigid metallic scaffold because injectable hydrogels cannot carry therapeutic molecules.
  5. Design the hydrogel to disassemble permanently as soon as it contacts water.

Correct Answer: 1.

Answer Rationale:
Self-assembly can form three-dimensional hydrogel networks through reversible non-covalent interactions under relatively mild conditions. This approach may support injectability, self-healing, and stimulus responsiveness, although the resulting material must still be tested for stability, mechanical strength, and biological performance under physiological conditions.

Reasoning / Explanation

Self-assembly is the most appropriate method because it can form a three-dimensional network under relatively mild conditions, reducing the risk of damaging the temperature-sensitive therapeutic molecule. Reversible interactions also allow the network to break and reform, which can support injectability, self-healing, and stimulus-responsive behavior.

Academic Principle / Supporting Evidence

The relevant principle is reversible supramolecular assembly through non-covalent interactions. The article identifies hydrogen bonding, hydrophobic interactions, π–π stacking, and electrostatic interactions as forces that can drive spontaneous hydrogel-network formation. It also explains that self-assembly offers mild preparation conditions and allows reversible deformation under changes in temperature, pH, or light.

Sample Question 1

Assessment Focus: Inferring relationships between interconnected urban services

—————————————–

The article explains that the effectiveness of healthcare services may depend partly on how well public transportation carries patients and healthcare workers to hospitals. Which conclusion is best supported by this example?

  1. Healthcare performance should be evaluated independently from all other city services.
  2. Public transportation creates value only when it generates direct financial revenue.
  3. The performance of one urban service may depend on supporting systems, so city planning should consider cross-service relationships.
  4. Hospitals should be responsible for operating all public transportation services.
  5. Digital health technologies can completely replace the need for physical transportation.

Correct Answer: 3

Sample Reasoning / Explanation:
Healthcare access does not depend only on hospitals or medical technology. Transportation can affect whether patients and employees reach healthcare facilities efficiently. Therefore, urban services should be assessed as interconnected systems rather than isolated operations.

Academic Principle / Supporting Evidence:
The example reflects systems thinking, in which the performance of one component depends on its relationship with other components in the wider urban ecosystem.

—————————————–

Sample Question 2

Assessment Focus: Understanding the purpose of a multi-stage literature-search process

—————————————–

Why did the researchers use multiple databases, synonyms, Boolean operators, and wildcard search terms before applying detailed screening criteria?

  1. To guarantee that every retrieved publication would be directly relevant to the research question
  2. To obtain broad initial coverage and then systematically remove duplicate, irrelevant, or ineligible studies
  3. To ensure that each database contributed exactly the same number of publications
  4. To avoid the need to assess the full text of the retrieved studies
  5. To include reports and publications that had not undergone peer review

Correct Answer: 2

Sample Reasoning / Explanation:
Broad search terms and multiple databases increase the likelihood of finding relevant studies expressed through different terminology. However, they also retrieve irrelevant or duplicate records, making systematic screening necessary.

Academic Principle / Supporting Evidence:
A systematic literature review balances search sensitivity—finding as many potentially relevant studies as possible—with screening specificity—retaining only studies that meet the defined eligibility criteria.

—————————————–

Sample Question 1

Assessment Focus: Assigning stakeholder roles in a knowledge-intensive smart-city project

—————————————–
A city plans to procure an AI-based public-health platform but lacks sufficient expertise to compare vendors, evaluate health evidence, and assess long-term social impacts. A local university offers to participate in the project.

Which arrangement would make the best strategic use of the university?

  1. Give the university complete ownership of the platform and authority over public-health policy.
  2. Use the university only to promote the project after the technology has been purchased.
  3. Engage the university as a knowledge intermediary, evaluator, and facilitator that reviews evidence, connects stakeholders, and assesses outcomes while the city retains public accountability.
  4. Ask the university to replace all government agencies and private technology providers.
  5. Restrict the university to collecting data without allowing it to examine project design or implementation.

Correct Answer: 3

Sample Reasoning / Explanation:
The city requires independent expertise and stakeholder coordination, but public authority should remain with the government. A university can support evidence evaluation, knowledge exchange, and project assessment without replacing the responsibilities of other actors.

Academic Principle / Supporting Evidence:
The article identifies universities as potential knowledge intermediaries, gatekeepers, providers, and evaluators in smart-city projects. Their role can strengthen informed decision-making and collaboration among government, businesses, technology providers, and communities.

—————————————–

Sample Question 2

Assessment Focus: Designing a real-world test for a factor that may function as both a benefit and a barrier

—————————————–
An agricultural cooperative believes that greater data transparency will improve trust among members. However, some farmers are concerned that sharing more information may create privacy risks and reduce participation.

Which approach would best test these competing expectations?

  1. Launch the platform across the entire region immediately and evaluate only the number of uploaded records.
  2. Ask the technology provider whether transparency normally improves trust.
  3. Conduct a phased pilot across several communities, measure participation and data use, and combine operational results with interviews or focus groups concerning trust, privacy, costs, and adoption.
  4. Select only the most technologically advanced farm and generalise its results to all agricultural communities.
  5. Review the number of transparency-related benefits in the literature without collecting evidence from users.

Correct Answer: 3

Sample Reasoning / Explanation:
Transparency may increase confidence when users understand how data are used, but it may also create concern when governance and privacy protections are unclear. A phased mixed-method pilot would reveal how these effects operate in different real organisational settings.

Academic Principle / Supporting Evidence:
The article explains that benefits and barriers are context-dependent and interconnected. It recommends testing the review findings through real-world methods such as workshops, interviews, and focus groups rather than assuming that a factor will produce the same outcome in every organisation.

Sample Question 1

Assessment Focus: Interpreting comparative evidence while considering both effect size and the amount of supporting research.

—————————————–
Table 3 reports a weighted average improvement of 35.2% for career advancement, compared with 12.3% for educational attainment. However, career advancement was examined in only six studies, while educational attainment was examined in twelve studies.

Which conclusion is most academically appropriate?

  1. Career advancement is definitively the most reliable benefit of Inclusionary Zoning because it has the largest percentage.
  2. Educational attainment should be ignored because its weighted average effect is smaller.
  3. Career advancement shows a larger reported average effect, but the smaller number of studies means the strength and generalisability of this finding should be interpreted cautiously.
  4. The two outcomes cannot be compared because weighted averages have no meaning in a systematic review.
  5. The findings prove that Inclusionary Zoning improves career advancement for every resident by exactly 35.2%.

Correct Answer: 3.

Reasoning / Explanation:
The reported effect for career advancement is larger, but it is based on fewer studies. A larger effect estimate does not automatically provide stronger or more generalisable evidence, especially when the evidence base is comparatively limited.

Academic Principle / Supporting Evidence:
Interpretation of research findings should consider both the magnitude of an effect and the quantity, consistency, and methodological quality of the supporting studies.

—————————————–

Sample Question 2

Assessment Focus: Interpreting variable intervention outcomes without making an unsupported universal conclusion.

—————————————–

Among individuals who received speech and language therapy, parents of 42% reported a “high” or “very high” impact, while 33% reported a “low” or “no” impact. The average rating was 3.2 out of 5.

Which interpretation is best supported by these findings?

  1. Speech and language therapy is equally effective for every individual with JNCL.
  2. Speech and language therapy has no educational value because some parents reported limited effects.
  3. The therapy may provide meaningful benefits for some individuals, but variation in outcomes indicates that support should be individualised and evaluated over time.
  4. The average score proves that therapy permanently prevents speech decline.
  5. Therapy should be offered only after oral communication has become unintelligible.

Correct Answer: 3.

Reasoning / Explanation:
The results indicate that some individuals experienced substantial benefits, while others experienced limited effects. The evidence therefore supports potential usefulness but not a universal or identical response.

Academic Principle / Supporting Evidence:
Variation in intervention outcomes supports person-centred planning and continuing formative assessment rather than assuming that one intervention will produce the same result for every learner.

—————————————–

Sample Question 1

Assessment Focus: Designing a multidimensional evaluation system for an Inclusionary Zoning programme.

—————————————–
A city reports that its Inclusionary Zoning programme is successful because it has produced 1,000 affordable units. However, the city has collected no information about resident retention, access to schools and employment, community participation, or social interaction.

Which evaluation strategy would provide the most complete assessment of social sustainability?

  1. Continue measuring only the number of affordable units because housing production represents every social outcome.
  2. Measure property values only because higher prices demonstrate community cohesion.
  3. Compare construction costs with private developments and exclude resident outcomes from the evaluation.
  4. Combine housing-production data with measures of resident retention, cross-income interaction, educational and employment access, community participation, and implementation conditions.
  5. Evaluate the programme only through developer satisfaction surveys.

Correct Answer: 4.

Reasoning / Explanation:
The number of affordable units measures policy output but does not establish whether the programme has achieved social integration, economic mobility, community stability, or access to opportunity. A complete evaluation must examine several outcome dimensions and the institutional and market conditions influencing them.

Academic Principle / Supporting Evidence:
Social sustainability is multidimensional. Policy evaluation should distinguish outputs, such as units produced, from longer-term social outcomes and should use evidence from several indicators to guide policy refinement.

—————————————–

Sample Question 2

Assessment Focus: Applying the concepts of person-centred transition planning and life flow to adulthood.

—————————————–
A 19-year-old student with JNCL is preparing to leave school. The adult service proposes a completely new daily programme based on available institutional activities, even though these activities are unrelated to the student’s long-standing interests, relationships, and previous routines.

Which alternative would best reflect the article’s concept of “life flow”?

  1. Accept the institutional programme because continuity is less important after leaving school.
  2. Design the transition around the individual’s history, preferred activities, relationships, skills, and interests, while adding the support needed to maintain meaningful participation.
  3. Remove all familiar activities so that the individual is encouraged to become fully independent.
  4. Allow professionals to select activities without consulting the individual or family.
  5. Focus exclusively on medical care and discontinue educational, social, and vocational participation.

Correct Answer: 2.

Reasoning / Explanation:
The concept of life flow emphasises continuity across life stages. Transition planning should recognise the person’s previous experiences, interests, social relationships, and preferred activities rather than replacing them with a standard programme based solely on service availability.

Academic Principle / Supporting Evidence:
Person-centred transition planning adapts services to the individual. It uses interdependence, environmental support, and collaboration to preserve autonomy, identity, continuity, and meaningful participation as support needs increase.

PART I | UNDERSTAND THE SCIENCE
Biomedical & Biopharmaceutical Scientific Understanding

Based on: Differential polyamine metabolism in CHO cell lines: Insights into cell growth and antibody quality

1 | SAMPLE QUESTION 1
Assessment Focus:
Assessment Focus: Interpretation of experimental results and application of cell-line differences to bioprocess design

English
The study found that removing putrescine (PUT) from the culture medium caused a substantial reduction in the growth of CHO-K1 cells, whereas CHO-DG44 cells were considerably less affected. Which conclusion is most appropriate for developing culture media for biopharmaceutical production?

  1. The same culture medium formulation should be used for all CHO cell lines to make experimental results easier to compare.
  2. Putrescine should be added at the highest possible concentration to all cell lines to maximize antibody production.
  3. Culture medium composition should be optimized according to the metabolic characteristics of each cell line.
  4. Only CHO-K1 should be used because it shows the clearest response to changes in culture medium composition.
  5. Polyamines should be avoided in antibody production because they make the process more complicated.
  • Correct Answer: 3
  • Reasoning / Explanation: The experimental results demonstrate that different CHO cell lines respond differently to PUT depletion. CHO-K1 showed a much greater reduction in growth than CHO-S and CHO-DG44. Therefore, using the same medium formulation for all CHO cell lines may not provide optimal process performance.
  • Academic Principle / Supporting Evidence: The article demonstrates that CHO cell lines differ in their polyamine requirements and metabolic characteristics. The authors therefore suggest that culture media should be optimized according to cell-line-specific characteristics to improve bioprocess efficiency and product quality.

2 | SAMPLE QUESTION 2
Product Quality
Assessment Focus: Analysis of the relationship between productivity and product quality

English
The study found that PUT depletion reduced antibody production but increased antibody galactosylation in some cell lines. If a researcher is developing a therapeutic antibody production process, which interpretation is most appropriate?

  1. PUT should always be minimized because galactosylation is more important than product yield.
  2. PUT should always be maximized because antibody yield is the only relevant measure of product quality.
  3. PUT is not important to the bioprocess because the cells can still produce antibodies.
  4. PUT levels should be optimized to balance cell growth, antibody productivity, and desired product quality attributes.
  5. Increased galactosylation confirms that every aspect of antibody quality has improved.
  • Correct Answer: 4
  • Reasoning / Explanation: PUT depletion does not produce only beneficial or only harmful effects. Although cell growth and antibody production decreased, some antibody glycosylation characteristics changed. Therefore, process development should not focus exclusively on either yield or product quality.
  • Academic Principle / Supporting Evidence: The article concludes that regulation of polyamine levels may be used as a strategy to modify therapeutic antibody glycan profiles. However, the balance among cell growth, recombinant protein yield, and desired glycan modifications must be carefully considered.

PART II | ANALYZE THE PROCESS
Bioprocessing & Biopharmaceutical Process Analysis

Based on: Purification of Monoclonal Antibodies Using Chromatographic Methods: Increasing Purity and Recovery

3 | SAMPLE QUESTION 3
Assessment Focus: Designing a purification sequence based on the separation mechanisms of different chromatographic techniques

  1. SEC → Protein A affinity chromatography → IEX
  2. Protein A affinity chromatography → IEX → SEC
  3. IEX → SEC → Protein A affinity chromatography
  4. HIC → Protein A affinity chromatography → Precipitation
  5. SEC → HIC → Protein A affinity chromatography
  • Reasoning / Explanation: Protein A affinity chromatography

English
A biopharmaceutical manufacturer is designing a downstream process for a monoclonal antibody containing an Fc region. The process has the following objectives:

  • Capture the antibody selectively from a complex mixture
  • Reduce charge-related impurities in the next step
  • Remove aggregates before obtaining the final product

Which sequence is most appropriate based on the principles described in the article?

  1. SEC → Protein A affinity chromatography → IEX
  2. Protein A affinity chromatography → IEX → SEC
  3. IEX → SEC → Protein A affinity chromatography
  4. HIC → Protein A affinity chromatography → Precipitation
  5. SEC → HIC → Protein A affinity chromatography
  • Correct Answer: 2
  • Reasoning / Explanation: Protein A affinity chromatography is suitable for the capture step because of its high specificity for the Fc region of monoclonal antibodies. IEX can subsequently separate molecules according to charge characteristics, while SEC can help remove aggregates or other species that differ in size from the desired antibody.
  • Academic Principle / Supporting Evidence: The article describes antibody purification using the general sequence of Capture → Intermediate Purification → Polishing. Different separation mechanisms can be selected according to antibody properties, impurity profiles, and required final purity. Protein A provides high antibody specificity, IEX separates according to charge, and SEC can remove aggregates and other size-related impurities.

4 | SAMPLE QUESTION 4
Recovery
Assessment Focus: Process decision-making based on the trade-off between purity and recovery

English
The article describes a cation-exchange chromatography development study in which one resin achieved nearly 100% monoclonal antibody purity, but only approximately 56.5% recovery. If the objective is industrial-scale manufacturing, which conclusion is most appropriate?

  1. The resin should be selected immediately because purity is the only important variable in biopharmaceutical manufacturing.
  2. IEX should be abandoned entirely because its recovery is below 100%.
  3. The trade-off between purity and recovery should be evaluated, and the resin, gradient, or operating conditions may require further optimization.
  4. Additional purification steps should be added without considering product losses.
  5. Recovery is not important as long as the final product meets the purity specification.
  • Correct Answer: 3
  • Reasoning / Explanation: Although nearly 100% purity is an excellent result, a recovery of 56.5% means that a substantial proportion of the product is lost during processing. Therefore, an industrial purification process should consider both product purity and the amount of product recovered rather than making a decision based on purity alone.
  • Academic Principle / Supporting Evidence: The article reports that cation-exchange chromatography performance can be affected by factors such as resin selection, elution mode, gradient slope, and flow rate. It specifically describes Nano SP-15L as achieving nearly 100% purity with 56.5% recovery. The article also discusses the trade-off between purity and recovery in purification process development, indicating the need to optimize process conditions according to the intended purification objective.

  1. Multiple-Choice Answer
  2. Reasoning / Explanation
  3. Academic Principle / Supporting Evidence

The examination does not assess only whether the selected answer is correct. It also evaluates the applicant’s ability to explain their reasoning, analyze scientific information, and support conclusions with appropriate academic principles or evidence. For each question, applicants should complete all three components:

  1. Multiple-Choice Answer Select the most appropriate answer from the five options provided.
  2. Reasoning / Explanation Explain why the selected answer is appropriate by connecting relevant data, experimental findings, or key information from the article to the situation presented in the question.
  3. Academic Principle / Supporting Evidence Identify the scientific principle, concept, study finding, or evidence from the article that supports the analysis and conclusion.

Guidance: A strong response should not simply state that “Option 3 is correct.” It should demonstrate which evidence leads to the conclusion, which scientific principle supports it, and why the selected answer is the most appropriate among the available options.
